Research Interest:

My research interests focus on the molecular mechanisms that control synthesis and crystallization of endotoxins produced by the entomopathogenic bacteria, Bacillus thuringiensis and B. sphaericus. Improving our knowledge of these aspects of endotoxin molecular biology is of both basic and applied value. A better understanding of the various genetic elements that control high levels of endotoxin synthesis is not only furthering our knowledge of protein synthesis in general, but is also contributing to the development of improved insecticidal bacteria. For example, by improving endotoxin yields per cell or per unit of fermentation medium, using recombinant DNA technology, I have made significant contributions to improving the insecticidal efficacy and cost-effectiveness of bacteria used in commercial insecticides. Similarly, a better understanding of the molecular basis of endotoxin crystallization could be useful for stabilizing insecticidal proteins, such as truncated Cry1 proteins, which do not crystallize, as well as for crystallizing other types of proteins for structural studies. To improve our understanding of endotoxin synthesis, my current research focuses on furthering our knowledge of the roles that several genetic elements play in this process, with an emphasis on the 5' STAB-SD and 3' stem-loop structures that stabilize mRNA, and the helper proteins active during and after translation. With respect to crystallization, my studies currently focus on identifying the regions responsible for crystallization in certain Cry proteins of B. thuringiensis and Bin protein of B. sphaericus.
